Utilizing our integrated technologies, Advanced Biotechnologies offers start-to-finish virus growth, purification, inactivation, and quality control testing services. Our technical staff will work closely with you when planning a production schedule suitable for your project. Working from well-defined protocols, we employ our experience in virology, mass cell culture, centrifugal separation and purification to successfully grow and purify high-quality custom virus preparations on a scale of 20 to 1000 liters.

Our large repository of cells, bacteria and viruses can be grown and purified as a custom order. You can choose what virus, product type and quality control testing you need.

An intended use form is required to be submitted with your custom quote request for projects using seed stocks from our list.

Growth

Advanced Biotechnologies can use our cell lines or a cell line you provide to grow high-quality cell seed stocks and create Master or Working Cell Banks (MCB or WCB). Our facility is set up to accommodate growths from 20 liters to 1000 liters of suspension cell culture and up to 500 liters of attached cell lines. Our cell culture team has experience with a variety of cell lines of insect, avian, and mammalian origin. Both suspension and anchorage-dependent lines are routinely cultured.

We can also use your provided virus stock to propagate your custom material.

Because Quality Control (QC) testing is a critical part of our production process, sterility testing, Mycoplasma testing, and a viable cell count are performed on every custom cell culture product. In addition, the following QC assays are available:

Purification

For more than 35 years, Advanced Biotechnologies has been purifying viruses using proprietary techniques, assuring the highest purity viral preparations. While all of our viral and bacterial preparations undergo rigorous quality control testing, the individual assays required to determine product specifications may vary according to your specific research needs. We provide multiple options to help researchers customize their viral and bacterial preparations.

Quality Control (QC) testing is a vital part of our production process. Sterility and Mycoplasma testing are performed at critical steps throughout the process. In addition, the following QC assays are available:

Inactivation

Inactivated viral and bacterial preparations are essential for some investigations. Advanced Biotechnologies offers custom inactivation services for any infectious agent in our inventory. Additionally, inactivations can be performed on samples submitted to us, or on custom viral or bacterial preparations.

Advanced Biotechnologies specializes in the following inactivation methods:

Beta-Propiolactone (ß-PL): A chemical inactivation method that modifies nucleic acid and leaves intact viral particles. ß-PL Inactivation causes minimal or no modification of viral proteins. Suitable for ELISA, Western blot, and other protein-based assays.

Detergent: A chemical inactivation method that leaves no viral particles intact. The detergent dissociates most viral proteins. Suitable for ELISA, Western blot, and other protein-based assays.

Heat: A physical inactivation method that leaves intact virus, bacteria, or chlamydia elementary bodies. The heat inactivates the infectious agents, but leaves the nucleic acid unmodified. Suitable for nucleic acid-based assays, such as standard and real-time PCR.

Ultraviolet (UV): A physical and chemical inactivation method that leaves intact viral particles. UV light cross-links the product’s nucleic acid with minimal or no modification of viral proteins. Suitable for ELISA, Western blot, and other protein-based assays.

Validation of inactivation, Sterility, and Mycoplasma testing are performed on every custom inactivation product. In addition, the following QC assays are available:

TEM Services

Looking for a full-service Electron Microscopy (EM) laboratory? We specialize in microbiology and virology applications on a cutting edge digital EM scope. For over 35 years our EM department has been serving the worldwide microbiology industry from start-up clients to the largest pharmaceutical companies. We will provide you with top quality images, fast turnaround time and a wealth of microbiology, virology and EM knowledge. Our facility also has the ability to accept many types of infectious biological samples.

Equipped with a Hitachi 7700 Transmission Electron Microscope, our technicians are able to detect biological samples as small as 20nm. Our high-quality images allow the examination of fine structure for the presence, absence, and quantification of known microbial or adventitious agents.

Imaging

We provide clients with digital photographs to aid in their quantitative or qualitative research efforts. These high resolution photographs are sent to the client in a digital format.

Particle Counts

Preparations to be quantitated for viral burden are prepared by mixing a known concentration of latex spheres with the unknown number of virus particles in the sample. After enumerating the sample for both virus particles and latex spheres, ratios are used to determine the concentration of virus particles per milliliter. Samples to be processed for virus particle counts can be concentrated by ultracentrifugation if necessary.

Intensive Search

Intensive searching of samples in TEM requires a slow and methodical visual screening of particles and biological specimens over large portions, if not entire surfaces, of prepared grids. These searches naturally yield a much higher amount of imaging per sample, which helps researchers access much more information than they would normally receive via particle counting. Digital photographs are provided along with a written report, including an interpretive summary of the search results obtained.

Ultra-Thin Section Microscopy

Ultra-thin sectioning is the only method in electron microscopy where investigators can observe cells and tissues at an ultrastructural level with high resolution. This service allows investigators to directly observe the effects of disease or experimental manipulations on cell and tissue ultrastructure.

Specimens submitted for ultra-thin section analysis are fixed in electron microscopy grade fixatives, dehydrated, infiltrated, and embedded in plastic resin. The embedded cells or tissues are then ultra-thin sectioned using high quality diamond knives, which allows for direct electron microscopic observation of intracellular and extracellular areas of the cells or tissues.

Client receives all digital photographs along with an optional detailed report in digital format describing materials and methods used and providing an interpretive summary.

FAQs

When does work on my TEM order start?
Work on your order begins once we receive the signed quote, purchase order, pre-payment (if required), samples and any other applicable materials.

How do I send my samples/specimens?
Biohazardous specimens must be packaged and shipped according to current shipping regulations. Validation data of inactivation must be provided along with inactivated specimens.

Can you handle all types of specimens?
We do not handle or process the following specimens:

  • Biosafety Level 3 specimens
  • Microbes and toxins on the Select Agents and Toxins Registry
  • Any other high-risk agents
