DNA Controls for PCR
ABI’s DNA controls can be used as positive controls in nucleic acid amplification reactions. Quantitated controls can also be used to generate standard curves for quantitative PCR assays.
ABI supplies virus infected cell DNAs, virus transformed cell DNAs, and quantitated DNAs. Non-quantitated DNAs are purified from appropriate starting material, and then the concentration is determined by fluorometry using PicoGreen® dye. Quantitated DNAs are prepared from purified viral or other organism DNA, then the copy number is determined by real time PCR using the Roche LightCycler®. Custom Nucleic Acid Preparations are also available.
ABI’s DNA controls are purified from one of six different sources
- Viral Particles
- Infected Cells: transduced or pathogen infected cell cultures
- Cultured Bacteria: pelleted bacteria from liquid, plate, or slant culture
- Transformed Cells: cells transformed by specific viruses
- Cultured Cells: cells used to propagate virus
- Cloned Virus
Virus infected cell DNAs are isolated from virus infected cells and contain cellular and viral DNA in unknown proportions. Virus transformed cell DNAs are isolated from cells transformed by human papillomavirus (HPV) type 16 or type 18 and contain HPV viral DNA integrated into the cell genome. Both virus infected and virus transformed DNAs are supplied at a DNA concentration (viral plus cellular DNA) of 50 ng/µL.
Quantitated DNAs for viruses, bacteria, and other organisms such as Toxoplasma gondii and Mycoplasma pneumoniae are also available. Quantitated DNAs are supplied in volumes of 250 µL at 1-2 x 104 DNA copies/µL (except for BK and JC polyomaviruses, which are supplied at 1-2 x 105 DNA copies/µL).
Note: Quantitation with different instruments and by different methods may not give the same copy number results. ABI’s DNA Controls are for research use only and are not for use in diagnostic assays.
