DNA Controls

ABI’s DNA controls can be used as positive controls in nucleic acid amplification reactions. The preparations from purified virus, virus-infected cells, bacteria, and mycoplasma allow the investigator to amplify or test any region of the genome which is present in natural infections.

ABI supplies virus infected cell DNAs, virus transformed cell DNAs as well as Quantitated DNAs for viruses, bacteria, and other organisms such as Toxoplasma gondii and Mycoplasma pneumoniae. DNAs are purified from appropriate starting material by SDS/proteinase K digestion, phenol/chloroform extraction and ethanol precipitation. The concentration of non-quantitated DNA is determined by fluorometry using PicoGreen® dye.

Virus infected cell DNAs are isolated from virus infected cells and therefore contain both cellular and viral DNA in unknown proportions. Virus transformed cell DNAs are isolated from cells transformed by human papillomavirus (HPV) type 16 or type 18 and contain HPV viral DNA integrated into the cell genome. Both virus infected and virus transformed DNAs are supplied at a DNA concentration (viral plus cellular DNA) of 50 ng/μl.

Quantitated DNAs are prepared from purified viral or other organism DNA and are supplied at 1-2 × 104 copies/μl (with the exception of BK and JC polyomaviruses which are supplied at 1-2 × 105 copies/μl). The copy number of Quantitated DNAs is determined by real time PCR using the Roche LightCycler. Quantitation with different instruments and by different methods may not give the same copy number results. These quantitated DNAs are for research use only and are not for use in diagnostic assays.

ABI’s DNA controls are purified from six different sources:

  1. Sucrose Density Gradient Purified Virus: virus that is sucrose banded
  2. Infected Cells: transduced or pathogen infected cell cultures
  3. Cultured Bacteria: pelleted bacteria from liquid, plate, or slant culture
  4. Transformed Cells: cells transformed by specific viruses
  5. Cultured Cells: cells used to propagate virus
  6. Cloned Virus

ABI’s Quantitated Nucleic Acid Control Information:

  • Each vial contains 250 μl total volume with 1-2 × 104 copies/µl
  • The copy number may vary depending on the method of quantitation
  • Quantitation is done by real time PCR using specific oligonucleotide primers
